唐高宗年间，阎立本偶遇狄仁杰（任嘉伦饰）蒙冤入狱事件，在助其自救后，发现他是断案奇才。适逢皇上派他去感业寺查案，遂决定带狄仁杰一同前往。在感业寺，狄仁杰侦破奇案，化解了一场佛门大劫，并与武媚娘结缘。狄仁杰一鸣惊人，被皇上派往并州，在此期间，他屡破奇案，被称为神判。时有前朝鬼兵杀人事件，狄仁杰前去查看，发现与前朝宝藏及濮王李泰意图造反有关，却被毁灭证据，无法继续。此时，发生武媚娘杀害亲女，嫁祸王皇后之事，皇上召狄仁杰回长安调查。根据武媚娘提供的线索，狄仁杰破获此案，并在李泰的住所找到前朝宝藏，成为濮王谋反的确实证据。而狄仁杰也知道此前破案过程中每遇瓶颈时为他解惑的人竟是武媚娘，只觉武媚娘深不可测，野心更是超乎自己的想象。果然不久，唐高宗因风疾发作，让武媚娘一同上朝，合称二圣临朝 。
